You have a 4L80-E and a 10.5" rear end, right? That drive train will handle anything you want to do.
Nearly everything available for the 5.3 will work on your 6.0 - same block.
Take your pick - turbo, twin screw supercharger, roots supercharger, centrifugal supercharger, nitrous oxide...
And of course heads, cam, headers, exhaust, etc...
Define your budget and you'll find some answers.

yeah everybody will go with FI right off the bat but start small and work your way up you might try stall, heads, cam , tune and see how you like that i will run really hard with that especially with all the cams you can pick from with your 6.0 not that i have a problem with FI its just an expensive start IMO but if you want to start big i say ATI/Vortech (centrifugal SC) or the a turbo kit as far as the STS i hear a lot of great stuff about them but i also hear some people prefer different things (A.k.a Parish8

i'm guessing this is your tow vehicle/daily driver. a good place to start if you are still all stock is a cat back, cold air induction kit, and a pcm reprogram. from there on a mild cam would do you well and if you want more torque, go radix.
