Throttle body coolant hose????

I wouldn't block it off unless you vent all of the air out of the heads before it is permanently blocked.
The hose is there to help vent air bubbles out of the heads which can lead to hot spots. Older gm motors and the 4.3 v6 do not require the hose because the upper radiator hose is in the intake manifold (highest point at which the coolant flows through the engine.) In the LS motors all of the coolant flows through the water pump which is much lower than the heads. Therefore air can be trapped in the top of the heads because air is lighter than water.
