At stock power level with stock everything else a tune really won't make much difference in how it tows that trailer.

Re-gear that truck if you want a change you will notice, then get a tune to make the transmission function correctly with those gears.

You need a tuner that adds about 5-7psi, 100+lbft of tq, gear swap, and better transmission.

All true, but I'd probably start with gears and a tune. 4.10 or 4.56.

Roots blower

I know I sound like a broken record, but its a proven combo, time and time again. cheap too. You could do it all for 3-3500 if you shopped around, got used stuff and got a good deal on gears and install. Deeper gears save the trans too.

Everyone saying you should Re-Gear is correct.
4.56 gears, and a PD blower will be your BEST upgrade for towing.
