87 or 91 Octane Dyno Tune for 00' 5.3?

IF this was a 2500 or 3500 truck used 70% of the time for towing heavy loads, then his advice might have been helpful buuuut it's not so either he didn't read the post or he doesn't care.

Everyone forgot to burn the OP on shorty headers. They are just like the pills that your mother gives you, they don't do anything at all. Go ask Alice, when she was 10 feet tall.
I don't think that you could do anything to improve gas mileage, short of something drastic. Like put a Gear Vendors overdrive. But you couldn't use it towing except maybe going downhill.
As an example, I have been towing a 20' ski boat for 14 years. When I pulled it with my truck with a supercharged 408 that got 14 hwy mpg empty, it got 12 mpg pulling the boat. When I towed it with my E150 with a 4.6 that gets 17 mpg hwy empty, , it got 12 mpg pulling the boat. When I pull it with my truck now with a LM7 with headers, that gets 16 mpg highway empty, , it got 12 mpg pulling the boat.
I don't know if this mpg information helps in any way, just keep it in the back of your head. But seriously, either install long tube headers, or leave the stock manifolds.

Needs a set 2.43 gears and the new BTR "Pee Wee Herman" camshaft (210/212 .450"/.450" 217*).
Set the rev limiter to 2,900 RPM's, lock the converter at 35 MPH and top speed fuel cutoff at 66 MPH.
Turning the key off and coasting down hills is bad on the 4L60e... so proceed with caution.

My "Tow Motor" is a gen3 4.8, 212/218 TSP .550"TSP ow lift, Magnuson tvs1900 with 3.2" pulley makes 11 PSI manifold pressure, 4.56 gears is the KEY for towing, let's the little mouse motor spin easy. Best Mo-Gas in the state is 90 octane at the pump.... 212,000 miles on the rig, been through 11 states, 4 provinces: Oil changes, tire rotations, air filters. So far nothing's broken.

My Tahoe runs 4.56 gears and 11 pounds boost, will pull ANYTHING with a 2-5/16 ball hitch..... or BREAK the trailer tongue off.
Three most important things for TOWING:
1) Gear ratio
2) Transmission cooling
3) Manifold pressure
