I have the following installed and run each one once a week.
1). SuperAntiSpyware (seems to catch more than Malwarebytes).
2). Malwarebytes - even though I'm running the above. Can't be too protective.
3). CCleaner - after you run the first two, run this to clean up your Registry. You'll see the option. This will clean up all registry entries leftover from uninstalls and other crap.
Make sure you back up the registry - you will be prompted to do so.
4). If you don't already have an Anti-Virus application, download the free version of AVG and set up an auto scan to run weekly when you're not on your computer.
Additionally, you may want to see what is in your startup folder and remove any unwanted applications - this will prevent them from starting up at boot time.
Oh and defrag, what ducky said above.
