im doing this swap also, and it is a plug and play deal as long as the 6.0 is either an lq4 or lq9, the newer ly6 is a gen IV 6.0 with variable valve timing and has the 58x reluctor wheel and the cam sensor is on the front instead of the back, also if its the 99-01 iron head 6.0 it will have a longer crank than the 02-06 6.0 so it wont work with the 4l60e unless you have a custom torque converter made.

That is to the best of my knowledge someone else will chime in im sure and correct me if i missed something.

As for a tune, its always a good idea to get a re tune done for the new motor but even if you don't the comp will make its own adjustments to handle it. it just may take a while for it to "relearn" it .

Only 99-00 had Iron Heads with the long crank. 01-up will have aluminum heads and the normal crank with spacer.

The stock 5.3 tune will have to be changed for the 6.0 engine to run correctly. It will most likely run lean. The computer may allow it to start and run but it doesn't know what injectors are on the actual engine or that the size of the cylinder has changed.

You should ask for help in the engine section and or the tuning section.
